چکیده

Photoacoustic microscopy (PAM) is a powerful imaging modality that can generate highresolution, three-dimensional (3D) images non-invasively. However, most types of cells cannot be imaged and quantified by PAM due to the lack of an absorption-based contrast mechanism. Here we demonstrate that this problem could be solved by employing 1-(4,5-dimethylthiazol-2-yl)-3,5diphenylformazan (MTT formazan), a cellular metabolite of 3-(4,5-dimethylthiazol-2-yl)-2,5diphenyltetrazolium bromide (MTT), as the contrast agent. The as-formed MTT formazan crystals inside the cells have a strong absorption in the spectral region of 500-700 nm, and are generally nontoxic. It could serve as a superb contrast agent for PAM in imaging various types of cells cultured on two-dimensional (2D) substrates and in 3D porous scaffolds. This contrast mechanism is general, and can be applied to essentially all types of metabolically active cells including stem cells and tumor cells, which would be very useful for a number of biological and biomedical applications.

Ultrahigh resolution photoacoustic microscopy via transient absorption

We have developed a novel, hybrid imaging modality, Transient Absorption Ultrasonic Microscopy (TAUM), which takes advantage of the optical nonlinearities afforded by transient absorption to achieve ultrahigh-resolution photoacoustic microscopy. The theoretical point spread function for TAUM is functionally equivalent to confocal and two-photon fluorescence microscopy, potentially enabling cell...

Noninvasive photoacoustic angiography of animal brains in vivo with near-infrared light and an optical contrast agent.

Optical contrast agents have been widely applied to enhance the sensitivity and specificity of optical imaging with near-infrared (NIR) light. However, because of the overwhelming scattering of light in biological tissues, the spatial resolution of traditional optical imaging degrades drastically as the imaging depth increases. Here, for the first time to our knowledge, we present noninvasive p...

Finite Element Simulation of Contact Mechanics of Cancer Cells in Manipulation Based on Atomic Force Microscopy

The theory of contact mechanics deals with stresses and deformations which arise when the surfaces of two solid bodies are brought into contact. In elastic deformation contact occurs over a finite area. A regular method for determining the dimensions of this area is Hertz Contact Model. Appearance of atomic force microscope results in introduction of Contact ...

Noninvasive Stem Cell Labeling Using USPIO Technique and their Detection with MRI

Background: To date, several imaging techniques to track stem cells are used such as positron emission tomography (PET), single photon emission computed tomography (SPECT), Bioluminescence imaging (BLI), fluorescence imaging, CT scan and magnetic resonance imaging (MRI). Although, overall sensitivity of MRI compared to SPECT and Bioluminescence techniques are lower, but due to high spatial reso...
